HPLC Purity vs Peptide Content

Why a high chromatographic purity result does not automatically mean the vial contains the same percentage of net peptide by mass.

Reviewed July 2026Buyer & quality briefingResearch supply context
Essential point

HPLC purity usually expresses the target peptide peak relative to peptide-related peaks under a defined method. Peptide content addresses how much of the material mass is peptide rather than water, counterions, salts or other non-peptidic components.

01 / Review framework

What procurement and laboratory teams should review

01

Identify whether the reported number is area-percent purity or quantitative assay

02

Check whether water and counterion contributions are measured separately

03

Review the reference standard and detection conditions used by the method

04

Avoid converting a purity percentage directly into net peptide mass

02 / Technical interpretation

Read the result in the context of its method

Analytical values become decision-useful only when the sample, batch, procedure, units, acceptance criterion and result belong together. A single headline percentage cannot answer identity, purity and content questions at the same time.

  • Confirm the document is lot-linked, not a generic example.
  • Distinguish qualitative identification from relative purity and quantitative assay.
  • Check method scope, reporting units and any stated exclusions.
01Define the material and form
02Match the batch and method
03Compare criterion with result
04Resolve exceptions before release
03 / Supplier discussion

Questions to resolve before quotation or release

  1. Is net peptide content or assay included in the specification?
  2. How were water and counterion levels accounted for?
  3. Which result should be used for quantitative laboratory preparation?
HK PEPTIDES project note

For wholesale projects, state whether the requirement is chromatographic purity, net peptide content, assay, or a combination. These requirements can change analytical scope and price.
